放线菌素D/TNF-α诱导小鼠胰岛微血管内皮细胞凋亡的研究

摘要 目的研究肿瘤坏死因子1型受体(TNFR1)在小鼠胰岛微血管内皮细胞(IMEC)上的表达以及肿瘤坏死因子α(TNF-α)对小鼠IMEC的损伤机制。方法用小鼠胰岛微血管内皮细胞系MS-1细胞进行实验,用免疫荧光法研究TNFR1在MS-1细胞上的表达;用四氮唑盐(MTT)比色法检测细胞毒作用;用流式细胞仪C膜联蛋白V-异硫氰酸荧光素(Annexin V-FITC)、碘化吡啶(PI)染色检测TNF-α对MS-1细胞凋亡的影响以及Ac-DEVD-CHO(caspase-3的竞争抑制剂)的保护效应。结果(1)MS-1细胞表达TNFR1。(2)发现TNF-α单独对MS-1细胞无明显作用(P>0.05)。TNF-α对放线菌素D(actinomycin D,ActD)致敏的MS-1细胞有明显的细胞毒作用,并且随着TNF-α浓度的增加,MS-1细胞活力逐渐降低。(3)Ac-DEVD-CHO可以明显拮抗ActD/TNF-α诱导的MS-1细胞凋亡,并且呈剂量效应关系。结论小鼠IMEC上有TNFR1的表达;TNF-α能诱导ActD致敏的胰岛内皮细胞发生凋亡,而Ac-DEVD-CHO则对这种细胞凋亡有明显的拮抗作用。
