摘要
目的:通过检测survivin和smac基因在多发性骨髓瘤(MM)患者中的表达,从而探讨其与多发性骨髓瘤(MM)病情发展及疾病预后的关系。方法:采用实时荧光定量RT-PCR技术检测29例MM患者和12例正常人骨髓细胞中survivin与smac的mRNA的表达水平。结果:survivin与smac mRNA在MM患者骨髓瘤细胞中阳性表达率分别为65.5%和75.9%,其mRNA的表达水平初治组较正常对照组明显升高(P<0.01),治疗缓解后(缓解组)其表达水平明显下降,与初治组比较差异亦有统计学意义(均P<0.05),在复发组中suivivin和smac mRNA的表达水平又复升高。二者在MM中的表达水平成正相关(r=0.924,P<0.01)。结论:survivin和smac基因的表达与MM发病及预后密切相关,可作为MM发病、复发及预后判断的有意义指标之一。
Objective To study the expression of survivin and smac mRNA in multiple myeloma (MM) and their clinical significance. Method The mRNA expression of survivin and smac gene in 29 MM patients and 12 healthy adults were detected by using real-time quantitative PCR assay. Result The positive mRNA expression rate of survivin and smae gene in the MM patients was 65.5% and 75.9%, respectively, and the mRNA expression levels of the two genes in newly diagnosed MM patients were significantly higher than those in the normal control(NC) (P 〈 0.01 ). However, the mRNA expression of the two genes decreased significantly in the complete remission patients than those of NC (P 〈 0.05). In the relapsed patients, the mRNA expression of survivin and smac gene increased again. The smac mRNA expression was positively correlated with that of survivin gene (r = 0.924,P〈 0.01). Conclusion The mRNA expression of survivin and smae gene is closely related with the occurrence and development of MM, which may he used as a prognostic indicator for this disease.
