三孢布拉霉菌中辅酶Q9的分离与鉴定

摘要 从三孢布拉霉菌菌丝体中分离提取辅酶Q,经硅胶柱和制备色谱纯化后纯度达98%。样品在Craven反应中显蓝色,紫外光谱扫描在275nm处有最大吸收,高效液相分析显示样品保留时间比辅酶Q10的略小。红外光谱谱图表明样品具有辅酶Q9的特征化学键和分子结构,电喷雾质谱(ESI/MS)确定了所分离的辅酶Q分子量[M+H]为795.8,以上结果表明,本实验室所保存的三孢布拉霉菌菌丝体内的泛醌存在形态是辅酶Q9,这为后续的发酵生产和药物合成研究奠定了重要的工作基础。 CoQ was isolated from Blakeslea trispora with a purity of 98 % after purification with a silica gel column and preparative HPLC. The sample gave a blue color in the Craven reaction and a UV absorption maximum at 275 nm. CoQ showed a shorter retention time in HPLC as compared to CoQ10. FTIR analysis showed the characteristic absorption peaks of CoQ9. The molecular weight which is 795.8 was determined by ESI/MS. The results of the above analyses indicate the presence of CoQ9 in Blakeslea trispora. This work has great potential for future use in fermentation and in drug synthesis.
