摘要
目的将靶向Survivin基因的siRNA导入大肠癌HT-29细胞中,并研究其对HT-29细胞生物学特性的影响。方法体外合成2条靶向人survivin基因的siRNA,并将其导入HT-29细胞,通过RT-PCR、流式细胞仪以及MTT法检测细胞内survivin mRNA的表达水平,细胞凋亡及增殖情况。结果HT-29细胞内survivin mRNA的表达水平降低,细胞凋亡率上升且肿瘤细胞增殖能力下降。结论靶向人survivin基因的siRNA能有效降低目的基因mRNA在大肠癌细胞HT-29中的表达,抑制肿瘤细胞的生长。
[Objective] To investigate the effect of survivin siRNA on the biologieal features of HT-29 eells. [Methods] Two pairs of surviving siRNA were designed and synthesized in vitro. HT-29 was transfeeted with survivin siRNA. RT-PCR, flow cytometer and MTT were used to test the expression level of surviving mRNA, cell apoptosis and proliferation. [Results] The expression level of survivin mRNA in HT-29 was reduced dramatically. The apoptosis of the cells inereased and the capacity of cell proliferation decreased significantly. [ Conclusion] SiRNA targeted at human survivin gene can suppress the expression of survivin mRNA in HT-29, therefore, suppress the growth of carcinoma cells.
