低电导钙激活的钾通道(rSK3,rIK)抑制露蜂房蛋白1(NVP(1))抗细胞增殖作用 被引量:1

摘要 目的研究rSK3,rIK通道在细胞增殖中的作用。方法用凝胶层析和液相色谱从露蜂房水提物中分离一种纯蛋白,并检测此蛋白的抗细胞增殖作用;转染;Southern blot检测rSK3,rIK基因;细胞计数检测SK通道在细胞增殖中的作用。结果从露蜂房水提物中分离一种纯蛋白,分子量为6.6 ku,命名为NVP(1),能抑制Hek-293细胞增殖。Southern blot检测转基因rSK3,rIK成功。转rSK3,rIK基因后,能消减NVP(1)抗细胞增殖的作用。结论rSK3,rIK促进细胞增殖。 Aim To investigate the effect of rSK3 and rIK anti-NVP (1) on inhibiting the proliferation of Hek-293 cells. Methods The effect of rSK3 and rIK anti-NVP ( 1 ) on Hek-293 cell proliferation was detected by cell count assay. Transfection and southern-blot were used to detect the rSK3 and rIK gene. Resultsr SK3 and rIK gene could prompt proliferation and inhibit the effect of NVP ( 1 ). Conclusions rSK3 and rIK channel can prompt proliferation of Hek-293. SK may be a novel target on cancer therapeutics.
