摘要
根据水蛭素 (HV3)的氨基酸序列 ,设计并合成了六对 (共 12条 )互补寡核苷酸链。经退火配对、连接成完整的 HV3编码基因 (2 2 8bp) ,并克隆在载体 p UC18中。通过α-互补及小量抽提质粒酶切鉴定筛得阳性重组克隆 ,其中 ,p UCH33经正、反向引物测序鉴定 。
According to the amino acid sequence of hirudin Ⅲ (HV3), a 228 bp gene coding for HV3 was assembled from six long oligodeoxyribonucleotide pairs which were synthesized on Beckman oligo 1000 DNA synthesizer. The gene was cloned in pUC 18 , and sequenced on ABI373A automated sequencer. The gene for HV3 in recombinant pUCH 33 had the correct nucleotide sequence.
