摘要
通过L25(53)正交试验设计,在M S基本培养基上添加不同浓度的2,4-D、6-BA、NAA三种激素.试验分别用直立型扁蓿豆的下胚轴和子叶作为外植体,探讨三种激素对直立型扁蓿豆愈伤组织诱导及分化的影响以及适合直立型扁蓿豆愈伤组织诱导和分化成苗的培养基.结果表明:6-BA对愈伤组织的形成有很大的影响;2,4-D,NAA对诱导愈伤的形成没有明显的作用.直立型扁蓿豆愈伤组织诱导的较优培养基组合是M S+2,4-D 0.5 m g/L+6-BA 1.0 m g/L+NAA 1.0 m g/L.同时,2,4-D对直立型扁蓿豆分化成苗的影响是最明显的,其次是6-BA,变化不明显的是NAA.愈伤组织分化成苗的较佳培养基组合是M S+6-BA 1.0 m g/L.
Based on the orthogonal design of L25 (5^3), three hormones with five different concentrations including 2,4-D, 6-BA,NAA were added into MS medium. Cotyledon segments and hypocotyls segments of Meiilotoictes ruthenica (L.)Sojak cv. Zhilixing were used as explants to explore effects of three hormones on callus induction and differentiation. At the same time, the optimal mediums for callus induction and differentiation can be determined. The results showed:6- BA was the most improtant for callus induction. But 2,4-D and NAA had an unobvious effect on callus induction. The optimal medium for callus induction of Melilotoides ruthenica (L.)Sojak cv. Zhilixing was MS+2,4-D 0.5 mg/L+6-BA 1.0 mg/L+NAA 1.0 mg/L. At the same time,2,4-D was more significant than 6-BA for differentiation, but NAA had an unobvious effect for differentiation. The optimal medium for differentiation of Melilotoides ruthenica (L.)Sojak cv. Zhilixing was MS+6-BA 1.0 mg/L.
