人精子经BWW获能5~6小时后,分别用己酮可可碱(PF)、孕酮(P)和A23187三种已知的获能及(或)顶体反应促进剂处理1小时,对实验和对照组精子用异硫氰酸荧光素标记的甘露糖化牛血清白蛋白(MFITCBSA)作探针标记精子甘露糖受体(MR)。结果表明,获能促进剂PF并不促进MR表达而P和A23187则有显著促进MR表达的作用。P对MRII型的促表达作用略强于对II型,而A23187则对II型的促进显著强于I型。认为MR的表达是一种依赖于钙离子的细胞生物学过程,并与顶体反应有密切关系。推测MR可能具有介导精子与卵膜融合的作用。
Objectives: To examine the effect of pentoxifylline, progesterone and A 23187 on expression of mannoseligand receptor (MR) of human spermatozoa.Design: Experimental study performed with donor spermatozoa.Setting: Andrology laboratory in Fujian Medical University.Subjects: Donor semen from proven fertile man.Main outcome measure: Percentage of spermatozoa labeled with MFITCBSA.Methods: The swimmingup spermatozoa were incubated in capacitating medium BWW for 5~6 hours at 37℃ and then treated, respectively, with three kinds of known capacitation and/or acrosome reaction activators: progesterone, calcium ionophore A23187 and pentoxifylline. After an hour, the mannoseligand receptors(MRs) of experimental spermatozoa and their controls were probed with fluorescent isothiocyanatelabelled mannosylated albumin (MFITCBSA, Sigma A7790) and visuallized by fluorescence microscopy.Results: Progesterone and A23187 improved remarkably MRs expression, while pentoxifylline did not. Progesterone enhanced the MRs expression of pattern II, slightly stronger than of pattern III, whereas A23187 enhanced pattern III much stronger than pattern II. MRs expression of pattern II,were detected on the area of acrosome cap, while that of pattern III, the receptors concentrated as a bar like narrow bend at the equatorial segment and post acrosome area.Conclusion: According to the results we propose that MR might play an important role in intermediate human fertilization, especially in spermoocyte membrane fusion.
