摘要 目的:检测脑源性神经营养因子(brain-derived neurtrophic factor,BDNF)在人牙髓干细胞中的表达,初步观察其对细胞增殖和分化的影响。方法:采用酶消化法分离培养人牙髓干细胞;RT-PCR检测细胞中BDNF mRNA表达;MTT和碱性磷酸酶活性检测观察其对细胞增殖分化的影响。结果:RT-PCR反应扩增出BDNF mRNA阳性产物;与对照组相比,25ng/mL BDNF可显著促进牙髓干细胞的增殖(P<0.05);100ng/mL时可显著提高碱性磷酸酶活性(P<0.05)。结论:BDNF可表达于人牙髓干细胞,对细胞增殖分化有一定促进作用。 AIM: To examine the expression of BDNF in human dental pulp stem cells(hDPSC) and the effect of BDNF on the proliferation and differentiation of human dental pulp stem cells in vitro. METHODS : Dental stem cells were isolated and cultured by a digestion method, and MTT colorimetric method was used to detect the close -effect of BDNF on the proliferation of hDPSCs. The differentiation effect was evaluated by alkaline phosphatase (ALP) activity assay. RESULTS: When the experiment group was compared with the control group, BDNF of 25ng/ mL stimulated the cell proliferation significantly (P 〈 0.05) and the ALP activities were obviously increased with BD- NF of 100 ng/mL(P 〈 0.05). CONCLUSION: hDPSCs can express BDNF mRNA while BDNF may play a role on the proliferation and differentiation of hDPSCs.
