消乳块胶囊抑制DMBA诱发大鼠乳腺癌及其机制 被引量:4

摘要 目的:观察消乳块胶囊对二甲基苯蒽(DMBA)诱发SD大鼠乳腺癌的影响及其机制。方法:50天龄Sprague-Daw ley系健康雌性未孕大鼠30只随机分为3组:模型组、治疗组和对照组。模型组和治疗组大鼠均先经两次DMBA(15mg溶于1.5mL芝麻油,间隔1周)灌胃造模,同时治疗组大鼠以消乳块胶囊0.4g/(kg.d)灌胃,持续给药,在第14周处死。大鼠乳腺组织及肿瘤组织均作常规病理,免疫组化检测ERα、ERβ、PR、AhR的表达,酶联免疫吸附法测定血浆E2、P、LH、FSH水平。结果:治疗组癌变率明显低于模型组(P〈0.05),分别是25.0%(2/8)和71.4%(5/7)。同对照组相比,ERβ、PR和AhR在治疗组和模型组中升高明显(P〈0.05~P〈0.01),ERβ在治疗组升高更为明显,同模型组相比,差异仍有显著性(P〈0.05)。血清激素测定显示,同对照组和模型组相比,E2、FSH和LH在模型组中升高明显(P〈0.05~P〈0.01);治疗组E2、FSH仍高于对照组(P〈0.05~P〈0.01)。结论:消乳块胶囊对DMBA诱导的大鼠乳腺恶性肿瘤有预防作用,其机制可能是通过ERβ介导的保护作用和调节体内激素水平来实现。 Objective:To investigate the anticancer action of XiaoRuKuai capsule on 9, l0 - Dimethy - lbenz(a) anthracene(DMBA) induced tumor in Sprague - Dawley rats. Methods :50 days old female Sprague - Dawley rats were randomly divided into 3 groups: negative control group ,model group, therapy group. The model group and therapy group were given DMBA by intragastric administration (15mg in 1.5mL sesame oil,twice,one week interval). At the same time, the rats in the therapy group group were given the XiaoRuKuai capsule[0.4g/( kg·d) ] by intragastric administration. The rats were killed in 14th week. The breast tissue or neoplasma were observed by routine pathology ( HE stain). Immuno- histochemical method detect the expression rate of ERα, ERβ, PR and AhR in tumor and mammary dysplasia tissue. The plasma levels (E2 ,P,LH and FSH) were detected by ELISA assay. Results:The mammary carcinogenesis rate was higher in the model group than in the therapy group. (P 〈0.05) ,71.4% (5/7) vs 25.0% (2/8). Compared to the control group,The expression rate of ER13,AhR,PR in the model and therapy group were remarkably higher( P 〈 0.05 ~ P 〈 0. 01 ) ,The expression rate of ER13 was higher than the model group(P 〈0.05). For the levels of E2 ,FSH and LH in the serum,we found that they were remarkably higher in the model group comparing the control and therapy group( P 〈0.05~P 〈 0.01 ). After we gave the XiaoRuKuai capsule, E2 and FSH levels had going down remarkably ( P 〈 0.05 ~ P 〈 0.01 ), but were higher than the control group yet ( P 〈 0.05 ~ P 〈 0.01 ). Conclusion : The XiaoRuKuai capsule has efficacy on preventing the mammary carcinogenesis induced by DMBA . The mechanism is probably associated with regulating ERβand the hormonal levels.
