摘要
目的:建立自体髓核移植致大鼠非压迫腰椎间盘髓核突出神经根性疼痛模型,以阿斯匹林为阳性对照药物,运用透射电镜观察方法探讨优化活血利水方对移植髓核、神经根超微结构的影响。方法:40只SD大鼠随机分为对照组、模型组、中药组、阿斯匹林组,建立大鼠自体髓核移植致腰5神经根性痛模型,造模后1周起各组分别予生理盐水1.82mL、生理盐水1.82mL、中药汤剂1.82mL、阿斯匹林1.82mL(0.1g.kg-1.d-1),每日1次,连续用药4周。给药后第3、7天测大鼠左后肢机械刺激痛阈值、第31天取移植髓核和左侧L5神经根作组织病理学电镜观察。结果:术后3、7天模型组、西药组及中药组大鼠左后肢的机械刺激痛阈比术前显著降低。超微结构显示:对照组组织结构正常,模型组明显异常,西药组及中药组显著优于模型组,中药组略优于西药组。结论:优化活血利水方可减轻髓核和神经根病理损害程度。
Objective: To design Non - compressive model study of experimental lumbar nucleus pulposus prolapse breeding nerve root pain in rats by transplantation of autologous nucleus pulposus, using aspirin as the positive control drug,to investigate the effects induced by Huoxue Lishui Fang on ultra structure of transplanted nucleus pulposus and nerve root through TEM. Methods: 40 SD( Sprague - dawley) rats were chosed. The rats were randomly divided into the control group, model group,the Chinese medicines group and the aspirin group. To design L5 nerve root pain model induced by transplantation of autologous nucleus pulposus in rats. A week later, 1.82mL NS, 1.82mL NS , 1.82mL Decoction of Chinese medicine and 1.82mL aspirin (0. 1g·kg^-1·d^-1 ) were given separately once a day. Medication for 4 weeks in a row. On the 3rd day and the 7th day after medecine was given, test of rats' left hind mechanical stimulation pain threshold was made. A month later, the left side of the transplanted nucleus and L5 root Histopathology were taken out for electron microscopy. Results : 3and 7 days after the surgical operation, The left hind mechanical stimulation of the model group, the group of western medicine and the Chinese medicine group were significantly lower than the pre - operative pain threshold. Ultra structure showed the ultra structure of the control group is normal, the model group was signifi- candy abnormal, Western medicine group and Chinese medicine group were significantly better than the model group, the Chinese medicine group was slightly better than the western medicine group. Conclusion: Optimized Huoxue Lishui Fang helped to relieve pathological damage of nucleus pulposus and nerve root .
