摘要
核转录因子-κB受体活化因子配体(RANKL)-核转录因子-κB受体活化因子(RANK)-骨保护蛋白(OPG)系统是近年发现的调节骨吸收的关键信号通路。RANKL与RANK结合在正常骨构建与骨改建中调节破骨细胞生成、活化和存留,在多种病理状况下增加骨吸收。OPG与RANK竞争性地结合RANKL,以防止骨组织的过度吸收。笔者分别就RANKL、RANK和OPG及其在骨构建与骨改建中的作用作一综述。
Receptor activator of nuclear factor-κB ligand(RANKL)-receptor activator of nuclear factor-κB (RANK) and osteoprotegerin (OPG) discovered in the mid 1990s is the key signaling system for the regulation of bone resorption. RANKL-RANK signaling regulates osteoclast formation, activation and survival in normal bone modeling and remodeling and increases bone resorption in a variety of pathologic conditions. OPG protects bone from excessive resorption by binding to RANKL and preventing it from binding to RANK. This article reviewed regulation of RANKL-RANK-OPG in bone modeling and remodeling.
