摘要
c-myc第一外显子在c-myc转录调节中起着重要作用。为了解c-myc第一外显子的生物学效应,探讨c-myc的转录调控机制,为c-myc表达增高性疾病的基因治疗提供依据,我们采用分子克隆技术,将1.3kb的人c-myc第一外显子片段经克隆载体pUC19换得适宜克隆位点,回收目的片段再反向导入逆转录病毒表达载体pLNCX,构建成人c-myc第一外显子反义载体pLNC-aM1。
xon 1 of cmyc plays a key role in cmyc transcription control. In order to investigate the biology effect of cmyc exon 1, determine the mechanism of cmyc transcription regulation, and then to provide evidents for the gene therapy of cmyc overexpression diseases, we used gene clone technology to construct 1.3kb human cmyc exon 1 fragment into cloning vector pUC19 to gain proper cloning sites, then recovery the target fragment and put into retrovirus expression vector pLNCX in reverse direction to construct human antisense cmyc exon 1 recombinant vector pLNC-aM1.
