摘要
目的探讨慢性缺氧对先天性心脏病患儿心脏缺氧诱导因子-1α(hypoxia-inducible factor-1α,HIF-1α)表达的影响。方法收集非青紫型先天性心脏病患儿16例、青紫型先天性心脏病患儿12例,体外循环前留取右心耳组织,采用半定量RT-PCR方法检测右心耳组织中HIF-1α mRNA水平,用免疫组织化学方法检测右心耳组织中HIF-1α蛋白的表达。结果青紫型先天性心脏病患儿心脏组织HIF-1α mRNA相对表达量与非青紫组比较差异无统计学意义(P>0.05),青紫型先天性心脏病患儿心脏组织HIF-1α蛋白表达量与非青紫组比较差异有统计学意义(P<0.001)。结论慢性缺氧上调先天性心脏病患儿心脏组织HIF-1α蛋白表达,该作用可能参与介导青紫型先天性心脏病患儿心脏慢性缺氧适应的分子机制。
Objective To explore impact of chronic hypoxia on hypoxia-inducible faetor-1α (HIF-1α) expression in heart of congenital heart disease children. Methods Right atrial tissue was excised from 16 children with acyanotie congenital heart disease and 12 children with cyanotic congenital heart disease during cardiac surgery. Gene expression of HIF-1α was determined by haft quantitative reverse transcription-polymerase chain reaction (RT-PCR) method. Protein expression of HIF-1α was investigated by immunohistochemistry. Results There was no significant difference between cyanotic and acyanotie children in HIF-1α mRNA expression (P 〉 0.05) , however, HIF-1α protein expression was significantly increased in hearts of cyanotic heart disease compared with that of acyanotic heart disease (P 〈 0.001 ). Conclusions Chronic hypoxia upregnlates HIF-1α protein expression in cardiac tissue from children with cyanotic congenital heart disease, which might be involved in the molecular mechanism of heart adaptation to chronic hypoxia.
