摘要
目的研究鼠伤寒沙门菌感染小鼠在免疫抑制下Th细胞因子的动态变化,探讨沙门氏菌感染的免疫机制。方法建立免疫抑制和感染的小鼠模型。在个不同时点处死各组动物,观察小肠、肝脏、脾脏病理改变。流式细胞仪测量脾脏Th1和Th2细胞(分别以IFN-γ和IL-4代表)。结果免疫抑制组病理改变较菌群失调组轻,IL-4明显增加,IFN-γ有一定的下降,Th1/Th2比值降低。结论免疫抑制造成过度的Th2型反应,感染扩散。
Objective To analysis immuno-suppression of interfereon-gamma and interleukin-4 in mice with Salmonella typhimurium infection. Method 24 BALB/c mice were orally infected with S. Typhimurium by a single dose and divided into 2 groups, including experimental group with immunosuppressive agents (EG) and untreated group (UG), respectively. The spleen, liver and small intestine were pathologically detected with microscope. The interferon-γ(IFN-γ) and interleukin-4 (IL-4) of splenocytes were measured with flow cytometry. Result The expression of IL-4 in EG was significantly higher than that in UG. The expression of IFN-γ in EG was significantly lower than that in UG. The pathological lesions in EG were serious compared with that in UG. Conclusion Immunosupperssion may induce excessively Th2 type immunological response to salmonella infection.
