中药复方对糖尿病大鼠血管内皮核因子和单核细胞趋化蛋白-1mRNA表达的影响 被引量:7

摘要 目的探讨中药复方糖脉平对2型糖尿病大鼠血管内皮核因子(NF-κB)和单核细胞趋化蛋白-1(MCP-1)mRNA表达的影响。方法将大剂量链脲佐菌素腹腔注射(STZ 60 mg/kg)造模成功的Wistar大鼠随机分为模型组、糖脉平组、二甲双胍组、玉泉丸组,每2周称1次体质量并剪尾测血糖,第6周处死取主动脉标本,采用免疫组化SAB法测定NF-κB及半定量RT-PCR法检测MCP-1 mRNA表达情况。结果造模后,大鼠血糖及NF-κB和MCP-1 mRNA的表达明显增高,经治疗后,糖脉平组及二甲双胍组NF-κB和MCP-1 mRNA的表达明显降低,与模型组比较有显著性差异(P均<0.01)。结论糖脉平能减少糖尿病大鼠NF-κB和MCP-1 mRNA的表达,减轻血管内皮局部炎症反应,阻断和降低动脉硬化的形成。 Objective It is to explore the effect of Chinese medicine compound Tangmaiping on the expression of blood NF - κB and monocyte chemoattractant protein- 1 (MCP- 1) mRNA in rats with type 2 diabetes. Methods The models of type 2 diabetic Wistar rats were established with high dosage(60 mg/kg) of streptozotocin(STZ) by intraperitoneal injection. Then successful model rats were gotten and randomly divided into model control group, Tangmaiping group, Metformin group and Yuquanwan group. The weight of the rats was measured and their tails were cut off to determine blood sugar in every two weeks. After six weeks the rats were killed and their aorta specimens were gotten to determine the level of NF - κB by SAB methods and the expression of mRNA MCP - 1 by semiquantitative reverse transcriptase (PCR). Results After making model, the blood sugar and the expression of NF- κB and MCP- 1 mRNA obviously increased. After treatment, the expression of NF- κB and MCP- 1 mRNA decreased significantly in Tangmaiping group and Metformin group. Compared with that in model control group, the differences were significant( P 〈 0.01 ). Conclusion Tangmaiping can decrease the expression of NF- κB and MCP- 1 mRNA, relieve the inflammatory reaction of blood vessel endothelium, and inhibit arteriosclerotic formation.
