摘要
目的采用基因表达谱芯片技术,分析老龄肾阳虚证的发生与基因表达异常间的联系。方法筛选肾阳虚证患者3名和正常对照者3名,采用美国Affymetrix公司的HG-u133Plus 2.0基因表达谱芯片,通过组间比较,筛选出老龄肾阳虚患者差异表达基因。结果芯片数据组间比较分析后获得34条差异表达基因,聚类分析得到6条有特征性地差异表达基因,主要涉及细胞周期和凋亡,肿瘤发生以及免疫。结论老龄肾阳虚证的发生涉及一系列免疫紊乱的复杂的过程,可能与个体的衰老存在密切关联。
Objective To explore the relevance between aged kidney - Yang deficiency syndrome and the differentially expressed genes by cDNA arrary. Methods Three kidney - Yang deficiency patients and three healthy people were selected as samples during the epidemiologic survey of the kidney - Yang deficiency syndrome. The differentially expressed genes were screened by applying HG -u133Plus2.0 of Affymetrix GeneChip. Results 34 common differentially expressed genes were obtained after comparison between patients and healthy peoples. 6 differentially expressed genes which are related to cell cycle, apoptosis, tumorigenesis and immunity were obtained by cluster analysis. Conclusion The aged kidney - Yang deficiency syndrome is related to the dysfunction of the immune system, may be closed related to body ageing as well.
