通心络超微粉对过劳叠加高蛋氨酸饮食条件下大鼠主动脉内皮保护的研究 被引量:1

摘要 目的探讨通心络超微粉对过劳叠加高蛋氨酸饮食大鼠主动脉内皮损伤的干预作用及其可能机制。方法健康雄性Wistar大鼠随机分为正常对照组、模型组、贝那普利组和通心络组4组,正常对照组喂饲普通饲料,模型组喂饲添加3%蛋氨酸的饲料并叠加力竭游泳,贝那普利组在模型组基础上饲以贝那普利(10mg.kg-1.d-1)灌胃,通心络组在模型组基础上饲以通心络超微粉(0.3mg.kg-1.d-1)灌胃,连续给药,于6周末应用酶法检测血浆中ET、PRA及AngII水平,比色法检测各组血清NO、MDA水平及SOD活性,免疫组织化学染色法检测主动脉组织中ACE、AT1R的表达。结果模型组ET、MDA水平均显著高于正常对照组(P<0.05),NO水平及SOD活性均显著低于正常对照组(P<0.01);模型组血浆PRA及AngII水平均显著高于正常对照组(P<0.01);模型组主动脉组织ACE、AT1R阳性表达均多于正常对照组(P<0.01);2个药物干预组ET、MDA水平均显著低于模型组(P均<0.01),SOD活性均显著高于模型组(P<0.05,P<0.01),AngII水平显著低于模型组(P均<0.01),通心络组NO水平明显高于模型组(P<0.05),血浆PRA水平明显低于模型组(P<0.05)。主动脉内皮组织中ACE、AT1R的阳性表达亦明显少于模型组(P<0.01)。结论(1)过劳叠加高蛋氨酸饮食对内皮的损伤机制可能与RAS的过度激活及氧化应激有关。(2)通心络超微粉可在一定程度上减轻上述病理损伤,可能与其抑制RAS过度激活、抗氧化有关。 Objective To explore the interventional effect of Tongxinluo supermicropowder on aortic endothelium in rats with defatigation combined with homomethionin diet.Methods Sixty Wistar rots were divided into 4 groups randomly: control group,model group,Benazepril treatment group and Tongxirduo superrnicmpowder treatment group. The control group was fed with conventional feeds, while all other groups were trained by fatigued swimming and fed with 3 percent homometh ionin diet. The Benazepril treatment group and the Tongxinluo treatment group were given suspension of Benazepril ( 10 mg· kg^- 1· d^-1 ) and Tongxinluo supermicropowder (0.3 mg·kg^- 1· d^-1 ) by intragaslrie administration on the basis of homoth ionin diet, respectively.After lasted for 6 weeks, the blood plasma ET, PRA and AngII were detected by enzymic method, and the levels of blood serum NO and MDA, the activity of SOD were observed by chromatometry, the expression of angiotensin-convertion enzyme (ACE), angiotensin II 1 receptor (AT1 R) was detected in the aortic tissue with Results After 6 weeks, the levels of blood plasma ET and MDA in model group were higher than that in control group ( P 〈 0.05) ; while the level of NO and the activity of SOD in model group were decreased compared with control group ( P 〈 0.01) ;and the level of PRA and AngII in model group was significantly higher than those in control group( P 〈 0.01);The positive expression of ACE and ATI R in model group were increased significantly compared with control group ( P 〈 0; 01). In the two interventional groups, levels of ET and MDA were significantly decreased compared with model group ( P 〈 0.01) ,the activity of SOD were obviously higher than that in model group ( P 〈 0.05, P 〈 0.01), and the levels of AngI/were significantly decreased compared with model group( P 〈 0.01 ). In Tongxirduo treatment group, the level of NO was higher than model group ( P 〈 0.05), while the level of PRA was lower ihan model group ( P 〈 0.05). The positive expression of ACE and ATl R in aortie endothehum were less than model group ( P 〈0.01).Conehmon (1) It suggested that defatigation combined with homomethionin diet could injury vascular endothelium, it may be related to the activated renin angiotensin system (RAS) and oxidative stress. (2)It provided that Tongxirduo supermicropowder could release the pathological injury, and it may be related to restraining the activated RAS and antioxidation.
