2型糖尿病局灶性脑缺血大鼠海马组织iNOS的表达及原花青素的干预

Effect of Procyanidin on Expression of Inos in Type 2 Diabetes Mellitus Rats with Focal Cerebral Ischemia

摘要 目的探讨2型糖尿病局灶性脑缺血大鼠脑组织中诱导型一氧化氮合酶(iNOS)含量的变化及原花青素(pro-cyanidin,PC)对其表达的影响。方法将30例大鼠随机分为假手术组、2型糖尿病单纯脑缺血组及PC低剂量组(50 mg/kg)、中剂量组(100 mg/kg)及高剂量组(200 mg/kg),于脑缺血后24 h进行HE染色,免疫组化法测定海马组织iNOS含量。结果2型糖尿病局灶性缺血组大鼠海马组织iNOS活性明显降低(P<0.01);PC低、中、高剂量组均不同程度降低iNOS活性(P<0.01),改善神经细胞的受损程度:其中以中、高剂量组明显(P<0.05),但中、高剂量组间无明显差异(P>0.05)。结论PC对2型糖尿病局灶性脑缺血大鼠能够降低海马组织iNOS的活性。 Objective To investigate the content of induced nitricoxide synthase in hippocampus tissue of type 2 diabetes mellitus rats with focal cerebral ischemia and effects of procyanidin (PC) on it. Methods Thirty SD rats were divided into sham - operation, model, low- dose PC, mid -dose PC and high- dose PC group. After ischemia for 24h, we performed hematoxylin and eosin (HE) staining; the expression of iNOS was examined by immunohistochemical SABC method. Results Compared with sham - operation group, the increased number of the positive iNOS cells in cerebral hippocampus of model group, the difference between the two groups was of significant ( P 〈 0. 01 ). Compared with model group, the PC groups possessed lower the positive cells of iNOS. The medium - dose and high- dose groups'effects were more obvious compared with the low- dose groups (P 〈 0. 05 ), but there was no difference between the two groups ( P 〉 0.05 ). Conclusions PC can reduce the activity of proeyanidin on expression of iNOS in type 2 diabetes mellitus rats with focal cerebral ischcmia.
