黄颡鱼雌雄差异的SRAP标记 被引量:19

摘要 采用序列相关扩增多态性(SRAP)分子标记技术,共使用140对引物进行了黄颡鱼性别表达特征性序列的筛选研究。结果显示:引物上下游组合F2R8扩增获得的雌雄间差异性序列稳定可靠,重复性强。根据GeneTools软件对该特征性DNA序列的相对表达量在雌雄间差异的统计分析结果,设定此DNA序列相对表达量(10%)作为鉴别雌雄黄颡鱼的界定参考指标,低于10%判定为雌鱼,高于10%判定为雄鱼。 A study was conducted to screen the sex-associated DNA sequence of Pelteobagrus fulvidraco using 140 pairs of primers with the purpose to identify the sex of each individual by the sequence-related amplified polymorphism (SRAP) molecular marker technique. Then a sex-associated DNA sequence of P. fulvidraco, which was amplified by primer pairs F2RS, was obtained, and the band pattern was sufficiently reliable, effective and repetitive. Furthermore, the calculation and analysis of DNA% raw volume of this special DNA sequence by Gene Tools software provides a new method to identify the sex ofP. fulvidraco. DNA% raw volume greater than 10 percent was used as the evaluation criterion for male, and DNA% raw volume less than 10 percent for female.
