羟乙基淀粉急性等容血液稀释对兔心肌缺血再灌注损伤的抗氧化作用 被引量:3

The antioxidative effect of acute isovolemic hemodilution with hydroxyethel starch on myocardial ischemia/reperfusion injury in rabbits

摘要 目的观察6%羟乙基淀粉(hydroxyethel starch,HES 130/0.4)急性等容血液稀释对兔心肌缺血再灌注损伤的抗氧化作用。方法21只家兔随机分为3组,每组7只:组Ⅰ(假手术组);组Ⅱ(缺血再灌注组);组Ⅲ(血液稀释组)。观察在急性心肌缺血45 min及再灌注180 min状态下血浆及心肌组织中过氧化物歧化酶(SOD)、磷酸肌酸激酶(CPK)、乳酸脱氢酶(LDH)活性及丙二醛(MDA)含量的变化。结果缺血及再灌注后,组Ⅱ血浆SOD活性进行性下降(P<0.05),MDA含量、CPK、LDH活性进行性升高(P<0.05);与组Ⅱ相比,再灌注后组Ⅲ血浆SOD活性升高(P<0.05),LDH活性降低(P<0.05)。组Ⅱ心肌组织各项指标在缺血区和非缺血区均有显著差异(P<0.01);与组Ⅱ缺血区相比,组Ⅲ缺血区心肌组织SOD、CPK、LDH活性均升高(P<0.05)。结论6%HES急性等容血液稀释对心肌缺血再灌注损伤有保护作用。 Objective To investigate the antioxidative effect of acute isovolemic hemodilution with 6% hydroxyetbel starch (HES 130/0.4) on myocardial ischemia/reperfusion injury in rabbits. Methods We investigated the activities of superoxide dismutase (SOD), creatine phosphokinase (CPK), lactic dehydrogenase (LDH), and the content of malondialdehyde (MDA) in plasm and rnyocardium after 45 minutes of ischemia and 180 minutes of reperfusion.21 rabbits were randomly divided into 3 groups with 7 in each one:group Ⅰ (sham group) ,group Ⅱ (ischemia/reperfusion group) and group Ⅲ (hemodilution group).Results After ischemia, the activity of SOD in plasm in group Ⅱ decreased( P 〈 0.01 ), the contentof MDA and the activities of CPK, LDH increased( P 〈 0,05) ,and all the parameters changed further after reperfusion( P 〈 0.05) ;after reperfusion, compared with group Ⅱ ,the activity of .SOD in plasm in group Ⅲ increased( P 〈 0.05), and the activity of LDH in plasm in group m decreased( P 〈 0.05). All the parameters in group Ⅱ differed significantly between the isehemic myocardium and the non-ischemic myocardium ( all P 〈 0.01 ) ; compared withgroup Ⅱ , the activities of SOD , CPKand LDH in the ischemic myocardium in group Ⅲ increased(P〈0.05).Conclusion Acute isovolemic hemedilution with 6%HES 130/0.4 could exert protective effect on myocardial ischemia/reperfusion injury.
