癌性营养不良小鼠肠黏膜损伤与炎症反应相关性研究 被引量:3

摘要 目的:探讨癌性营养不良小鼠肠黏膜损伤与炎症反应的关系。方法:将14只小鼠随机分为对照组和荷瘤组。荷瘤组皮下种植小鼠结肠腺癌26细胞悬液(1×106个/只)。接种后检测两组小鼠的体质量变化。第16天测量小肠黏膜的绒毛高度、绒毛宽度、基底层厚度和肌层厚度,检测血清和小肠组织中肿瘤坏死因子-α(TNF-α)、IL-1、IL-6、IL-10、γ干扰素(IFN-γ)、分泌型肿瘤坏死因子受体Ⅰ(sTNF-RⅠ)和sTNF-RⅡ的量。结果:实验第16天荷瘤组小鼠去瘤体质量、小肠绒毛高度、基底层厚度明显低于对照组,有显著性差异(P<0.05)。绒毛宽度、肌层厚度无明显改变。荷瘤组小鼠血清中TNF-α、IL-6、sTNF-RⅠ浓度高于对照组,IL-10浓度则低于对照组。荷瘤组小鼠小肠组织中TNF-α浓度高于对照组,而IL-1、IFN-γ、sTNF-RⅠ和sTNF-RⅡ浓度均低于对照组。结论:癌性恶病质小鼠存在肠黏膜损伤,血清和肠组织中存在广泛的细胞因子浸润,两者可能有相关性。 Objective: To study the correlation between intestinal mucosa injuries and inflammatory reaction in cancer cachexia mice. Methods: 14 male BALB/c mice were randomized into two groups: control group and tumor-beating group. For tumor-beating group, a suspension of 10^6 murine colon 26 adenocarcinoma cells was inoculated s. c into each mouse. Non-tumor body weight, intestinal villus height, width, basal layer thickness and muscular layer thickness, TNF-α, IL-1, IL-6, IL-10, IFN-γ, sTNF-R Ⅰ and sTNF-R Ⅱ in the serum and intestinal tissue were determined at the 16th day following the inoculation. Results: The non-tumor body weight and intestinal villus height and basal layer width in tumor-bearing group were lost significantly as compared with control group. The concentration of serum TNF-α, IL-6, sTNF-R Ⅰ were higher and the level of serum IL-10 was less in tumor-bearing group. The concentration of intestinal tissue TNF-ct was higher but the IL-1, IFN-γ, sTNF-R Ⅰ and sTNF-R Ⅱ were less in the tumor-bearing group. Conclusion : The intestinal mucosa injuries have close relation with the inflammatory reactions in cancer cachexia.
