摘要
目的:探讨血红素氧合酶-1在LPS诱导急性炎症中的表达及臭苜蓿根提取物的干预作用。方法:通过脂多糖(LPS)干预RAW264.7细胞系建立炎症细胞模型。实时荧光定量RT-PCR法检测血红素氧合酶-1(HO-1)的基因表达差异,Westernblot法测定HO-1的蛋白表达量。结果:臭苜蓿根提取物明显升高巨噬细胞HO-1的mRNA表达水平,Westernblot结果显示,臭苜蓿根提取物干预后HO-1的蛋白表达水平也明显升高。结论:臭苜蓿根提取物能提高巨噬细胞HO-1基因的表达,促进HO-1的释放而发挥一定的抗炎作用。
Objective: To study the express of HO-1 on acute inflammation induced by LPS and the anti- inflammatory effect of the melilotus suaveolens extract. Methods : Inflammatory cell model was constructed by LPS acting RAW264.7 cell line. The expression of mRNA and protein of Hemeoxygenase 1 ( HO-1 ) were detected by real-time PCR and Western blot method. Results : The expression of HO-1 mRNA in macrophage cell was significantly enhanced by the melilotus suaveolens extract. The result of Western blot showed that the expression of HO- 1 protein was significantly increased after the intervention of melilotus suaveolens extract. Conclusion : The melilotus suaveolens extract can resist inflammation by enhancing the expression of HO-1.
