脂多糖对脐血CD34^+细胞体外增殖的影响

摘要 目的:研究脂多糖(LPS)对脐血干/祖细胞体外增殖的影响,探寻脐血干/祖细胞体外扩增的新方法。方法:将不同浓度的自制脂多糖和标准脂多糖分别加入含有脐血CD34+细胞的液体培养体系中培养,采用MTT测定方法分别测定脐血CD34+细胞增殖率。结果:不同浓度的自制脂多糖与标准脂多糖均可出现低浓度促进脐血CD34+细胞增殖,高浓度抑制其增殖的作用。最佳效应浓度分别在30U/mL及10mg/L。结论:脂多糖对脐血CD34+细胞体外增殖可能具有调节作用。 Objective: To study the effect of lipopolysaccharide (LPS) on cord blood hematopoietic stem/progenitor cells proliferation in vitro, and explore the new way of proliferation in vitro. Methods: Selfmadelipopolysaccharide and standard-lipopolysaccharideat different concentrations were added into cord blood CD34^+ cells liquid culture system respectively, the cord blood hematopoietic stern/progenitor cells was analyzed by MTT. Results: Selfmade-lipopolysaccharide and standard-lipopolysaccharide at different concentrations could promote proliferation of cord blood CD34 ^+ cells in LPS at low concentration, and were inhibited in high concentrations LPS. The best concentration were 30U/mL and 10mg/L respectively. Conclusion : Lipopolysaccharide may play a role in regulating the cord blood hematopoietic stem/progenitor cells proliferation in vitro.
