跨损伤DNA合成通路在肿瘤发生中的作用及其与化疗敏感性的关系 被引量:3

摘要 跨损伤DNA合成(translesion DNAsynthesis,TLS)属于一种复制后修复过程,主要包括DNA聚合酶κ、η、ι、ζ。跨损伤DNA合成途径是生物体的一种应急机制,能够在DNA损伤未被修复的状态下进行复制延伸,维持基因组稳定性,但也不可避免的会引起DNA突变。跨损伤DNA合成通路基因的上调或下调,均是促进肿瘤发生的潜在因素。此外,跨损伤DNA合成途径在修复铂类化疗药物引起的DNA损伤中发挥了主要作用。采用反义RNA或RNA干扰抑制DNA聚合酶ζ、η等表达后,肿瘤细胞对铂类化疗药物的耐受都明显下降,这为铂类药物的化疗增敏提供了新的思路和方法。
