肾透明细胞癌组织中转化生长因子β1和磷酸化细胞外信号调节激酶的表达

摘要 目的:探讨肾透明细胞癌(RCCC)组织中转化生长因子β1(TGF-β1)和磷酸化细胞外信号调节激酶(p-ERK)的表达及其临床意义。方法:采用免疫组织化学SP法检测72例肾透明细胞癌组织和30例正常肾组织中TGF-β1和p-ERK蛋白的表达,分析其与RCCC临床病理特征的关系。结果:①RCCC组织中TGF-β1和p-ERK阳性率分别为70.2%和78.1%,均高于正常肾组织22.6%和16.7%(P均<0.05),2者的表达呈正相关(r=0.443,P<0.05)。②TGF-β1和P-ERK的表达与RCCC组织分化程度和TNM分期有关(P<0.05)。结论:TGF-β1和p-ERK可能在肾透明细胞癌的发生发展过程中发挥重要作用。 Aim : To investigate the expression of transforming growth factor β( TGF-β) and phosphorylated activated extracellular signal-regulated kinase(p-ERK) and their clinical significance in renal clear cell carcinoma(RCCC). Methods:The expression of TGF-β1 and p-ERK was detected in 72 cases of RCCC tissue and 30 cases of nomal renal tissue by immunohistnchemical method. Results: The positive rates of TGF-β and p-ERK in RCCC were 70. 2% (50/72) and 78.1% ( 56/72 ) , respectively, higher than those in normal renal tissue [ 22.6% ( 7/30 ) and 16.7 % ( 5/30 ) ] , respective-ly. The expression of TGF-β1 was correlated with that of p-ERK(r = 0. 443 ,P 〈 0. 05 ). The positive rates of the TGF-β1 protein expressions were 83.3% (30/36) in poorly differentiated carcinoma,79.6% (43/54) in the Grade Ⅲ - Ⅳ carcino- ma, higher than that [ 55.6% (20/36) ] in well-moderately differentiated carcinoma, and that [ 38.9% (7/18) ] in the Grade Ⅰ -Ⅱ carcinoma, and there were significant differences among them ( P 〈 0. 05 ). The positive rates of the p-ERK protein expressions were 94.4% (34/36) in poorly differentiated carcinoma,88.9% (48/54) in the Grade Ⅲ- Ⅳ carcino- ma, higher than that [ 61. 1% (22/36) ] in well-moderately differentiated carcinoma, and that [ 44.4% ( 8/18 ) ] in the Grade Ⅰ-Ⅱ carcinoma,and there were significant differences among them(P 〈 0.05). Conclusion :The findings demonstrate that there are the abnormal expression of TGF-β1 and p-ERK in RCCC, and the positive rates of the TGF-β1 and p- ERK protein expressions are correlated with the grades and the stages of RCCC, which suggests that the two may contribute to the pathogenesis and progression of RCCC.
