VEGF在小鼠早期急性肺损伤中表达的变化 被引量:2

Changes of VEGF in the Acute Lung Injury in the Early Stage in Mice

摘要 目的观察油酸致小鼠早期急性肺损伤(ALI)中血管内皮细胞生长因子(VEGF)表达的变化。方法48只BALB/c小鼠随机分为两组,每组24只。ALI组从尾静脉注射油酸0.9ml/kg制备ALI模型;正常对照组注入等量生理盐水。8h后每组随机抽半数小鼠处死后行肺泡灌洗(BAL),另一半小鼠右肺称重后烘干,左肺留取病理标本;分别测定肺湿/干质量比、肺泡灌洗液(BALF)总细胞计数、中性粒细胞计数、蛋白含量变化;ELISA测定血清和BALF中白细胞介素6(IL-6)、VEGF的含量;免疫组化法检测组织中VEGF表达的变化。结果ALI组肺湿/干质量比、BALF中蛋白含量和总细胞计数、中性粒细胞计数、血清和BALF中IL-6含量、血清VEGF含量与正常对照组比较均明显升高,ALI组BALF中VEGF的含量和肺组织中VEGF表达积分吸光度值(IA)较正常对照组均明显降低,以上比较的差异均有统计学意义(均P<0.05)。结论小鼠早期ALI血清VEGF表达升高而BALF和肺组织中VEGF表达降低。 Objective To investigate changes of vascular endothelial lung injury(ALI) induced by oleic acid in the early stage in mice. Methods mice were randomly divided into 2 groups(24 each). The animals in normal saline (0.9 ml/kg); in the ALI group ALI was induced by oleic acid (0.9 ml/kg) intravenously. Then each group was randomly divided into 2 subgroups (12 for each). One Group:protein level, counts and differential in brochoalveolar lavage fluid (BALF) were detected. The levels of IL-6 cell and VEGF in both serum and BALF were measured by enzyme-linked-immunosorbent assay(ELISA). Another Group:wet/dry weight(W/D) of right lung was recorded. Pathological changes of left lung and VEGF ex- pressions in lung tissue were examined. The expression of VEGF was immunohistochemically detected in lung tissues as integrated optic density (IA). Results W/D, protein level, total cell number and classification of polymorphonuclear leukocytes (PMN) in BALF,IL-6 level in both serum and BALF, VEGF level in serum were significantly higher in ALI group compared with those in the control group(P 〈0.05), hut VEGF level in BALF in ALI group was significantly lower than that in the control group(P〈 0.05). The expression of VEGF described by IOD in the control group was significantly higher than that in ALI group in the lung tissue (P〈0.05). Conclusion It is suggested that VEGF expression in BALF and the lung tissue increased in ALI induced by oleic acid in the early stage in mice, but decreased in serum
