高糖对人肾小球系膜细胞分泌单核细胞趋化蛋白1及层黏连蛋白的影响 被引量:2

摘要 目的通过体外试验研究高糖对人肾小球系膜细胞(HMC)分泌单核细胞趋化蛋白1(MCP-1)和层黏连蛋白(LN)的影响。方法用高糖干预体外培养的HMC,以ELISA法检测细胞培养上清的MCP-1和LN水平。结果高糖组培养上清在不同时点(24,48,72h)MCP-1和LN的表达水平明显高于正常糖组和甘露醇组(P<0.05)。高糖干预24h后,培养上清MCP-1和LN水平开始升高,于72h达峰值,正常糖组和甘露醇组未见显著升高(P<0.05)。结论高糖呈时间依赖方式增加HMC对MCP-1和LN的分泌,而正常糖和甘露醇无此作用。MCP-1和LN的增加是由于高糖而非高渗所致,提示高糖所致的炎性反应可能参与糖尿病肾病的发生和发展。 Objective To investigate the effect of high glucose on monocyte chemotactic protein-1 (MCP-1 )and laminin (LN)in the human mesangial cells(HMC) in vitro. Methods Human mesangial cells were cultured under high glucose,the level of MCP-1 and LN in supernatant were measured by ELISA. Results The level of MCP-1 and LN in high glucose group at different time (24 h, 48 h and 72 h) was significantly higher than in the normal glucose and mannitol group (P 〈 0.05).Compared with the normal glucose and mannitol group,the level of MCP-1 and LN in high glucose group began to increase at 24 h and reached the peak at 72 h after intervention with high glucose (P 〈 0.05 ).Conclusion High glucose promote human renal mesangial cells secreting MCP-1 and LN by time dependency. Normal glucose and mannitol did not have this effect, which demonstrate that high glucose cause the Level of MCP-1 and LN increasing, not hyperosmosis, so inflammatory reaction caused by high glucose may play a role in the progression of diabetic nephropathy.
