摘要 以胶质芽孢杆菌L-k和大豆根瘤菌HH103为试验用菌株,在混合培养条件下采用摇瓶发酵法,探讨接种顺序、接种比例、初始pH值、装液量等因素对L-k芽孢生成量的影响。进一步对各单因素进行不同正交试验,得到了各因素的较好组合为:混合接种比例为1:1,初始pH值为6.5,装液量为250mL三角瓶装30mL培养液,接种顺序为先接种L-k之后4h接种HH103,这个组合下芽孢生成量达到2.43×108cfu·mL-1,比对照提高16.3%。 Bacilluse mucilaginosus L-k and soybean rhizobium HH103 were used as trial strains.In the same system of mixed culture, the effect of some important factors such as proportion of mixed inoculation,order of inoculation,medium volume and initial pit on spore measures of L-k was investigated by shaking culture.The result showed that the optimum conditions of culture were as follows: the proportion of mixed inoculation 1:1 (L-k:HH103), initial pH 6.5,medium volume 30mL in 250mL triangular flask, inoculating L-k 4 hour earlier than HH103.The spore measures were 2.43×10^8cfn .mL^-1, increasing 16.3% compared with CK.
