氯化锂-毛果芸香碱点燃癫大鼠模型中腺苷激酶表达的动态改变 被引量:5

The Dynamic Change of Adenosine Kinase in the Model Kindled by Lithium Chloride-Pilocarpine

摘要 目的:观察氯化锂-毛果芸香碱慢性点燃大鼠在点燃过程中脑内腺苷激酶(ADK)表达的动态演变情况,探讨癫发作与ADK的关系及时间演变特征,以评价ADK在癫发病机制中的作用。方法:氯化锂-毛果芸香碱点燃大鼠模型,分模型组、对照组和正常组。采用RT-PCR及Western blot的方法监测发作后24h和1、6个月时大鼠脑内ADK表达的动态改变情况。结果:氯化锂-毛果芸香碱点燃后24h时,RT-PCR及Western blot检测ADK的表达均低于正常对照组,差异有统计学意义(P<0.05或P<0.01);在点燃进入慢性期后的1和6个月时则相反,ADK表达较点燃前明显增高,差异有显著统计学意义(P<0.01)。ADK表达6个月时较1个月时更高(P<0.05或P<0.01),即慢性期ADK的表达增高随时间延长越加明显。结论:ADK在点燃过程中呈现双向改变特征,急性期表达降低,促进细胞外腺苷浓度的增高;进入慢性期,反复的癫发作导致了ADK的过表达,这可能是导致癫反复发作而无法控制的重要因素。 Aim: To determine the relationship between seizure and the dynamic change of adenosine kinase (ADK) and analyze the feature of this dynamic change. Methods: The animals were divided into three groups, a model group, a control group and a normal group. The normal group can help to determine if the aging of the animals can result in the change of ADK expression. Status epileptic rats were kindled by lithium chloridepilocarpine and the detection pionts was at the 24th hour, 1 st month and 6th month. The dynamic change of the expression of adenosine kinase was tested by RT-PCR and Western blot technique. Results: In the acute phase that is at the 24th hour after kindling, the expression of ADK was downregulated. The results of RT-PCR and Western blot all showed statistic difference compared with the control group (P〈0.05 or P〈0.01). While as time went into the chronic phase that is at the 1st and 6th month after kindling, the expression went to the opposite direction. The expression of ADK was upregulated. There are significant statistical difference compared with the control group (P〈0.01). The results of comparison of the 1 st and 6th month indicated that the expression of ADK was even higher in the 6th month than in the 1 st month. There are statistical difference (P〈0.05 or P〈0.01). The results suggested that the changes of the expression of ADK tend to be more and more obvious as time went by. Conclusion: The dynamic change of ADK during kindling presented the bidirectional change. In the acute phase, the expression was downregulated to impulse the secretion of adenosine, while the chronic and repeated seizure result in the change of ADK to the opposite direction. The over expression of ADK might be an important cause for the uncontrolable seizure finally.
