摘要
目的:探讨胰岛素样生长因子-1(IGF-1)在大鼠肝纤维化发生中的作用机制。方法:对32只雄性SD大鼠皮下注射40%四氯化碳(CCl4)油剂制备大鼠肝纤维化模型,分别于注射后1,4,8周处理动物,留取肝组织,应用免疫组织化学方法检测大鼠肝组织中IGF-1的表达。结果:免疫组化结果显示,IGF-1主要表达于肝细胞胞质中。注射CCl4诱导后,大鼠肝组织中IGF-1表达较正常对照明显增强(P<0.01,<0.05),且1,4,8周肝组织中IGF-1的表达强度呈明显递增趋势(P<0.01,<0.05)。结论:在大鼠纤维化肝组织中,IGF-1主要表达于肝细胞胞质,可能通过促进肝细胞和肝星状细胞增殖、活化而发挥促纤维化作用。
OBJECTIVE To explore the role of Insulin-like growth factor-1 (IGF-1)in liver fibrogenesis via investigating the expression of IGF-1 in rats with liver fibrosis. METHODS Liver fibrosis models of rats were made by subcutaneously injecting with CCh. Thirty two male SD rats (weight 250- 300 g) were randomly sacrificed 1,4 and 8 weeks after injection of CCh respectively,and their liver tissues were used for examining IGF-1 by immunohistoehemistry stain. RESULTS The expression of IGF-1 in rats after injection with CC14 was found chiefly in hepatocytes and all significantly higher than those in normal rats(P〈0. 01). Moreover,it increased progressively the expression of IGF-1 respectively at 1st, 4th and 8th week after injection with CCl4 (P〈0. 01)CONCLUSION The overexpression of IGF-1 may accelerate the procedure of liver fibrosis via enhancing hepatocytes and hepatic stellate cells proliferation.
