摘要
本研究利用体外淋巴细胞培养和噻唑蓝(MTT)比色分析法,检测了两种浓度多巴胺(DA,10-5和10-6mol/L)对大鼠肠系膜淋巴结细胞由刀豆素A(ConA)诱导的增殖反应的影响;并运用高效液相色谱(电化学检测法(HPLC-ECD),测定了大鼠在注射抗原绵羊红细胞(SRBC)后的2~7天,胸腺和脾脏中DA含量的变化。结果说明:(1)10-5和10-6mol/LDA都能显著增强T细胞增殖。(2)大鼠在体液免疫应答的第6天,胸腺和脾脏内的DA含量显著增多,其它时期DA含量虽有变化,但无显著意义。以上结果提示,外周DA既可增强细胞免疫功能。
In this study, we determined, in vitro, the effect of dopamine (DA, 10-5 and 10-6 mol/L) on the proliferation of the cells in the rat mesenteric lymph nodes induced by concanavalin (Con A) and sought the change of content of DA in the thymus and spleen of rats immunized with sheep red blood cells (SRBC) during day 2-7 post-immunization by the aid of the high-performance liquid chromatography with electrochemical detection (HPLC-ECD). The results are as follows: DA at 10-5 and 10-6 mol/L significantly enhanced the T lymphocyte proliferation. On day 6 of humoral immune response, DA content in the thymus and spleen markedly increased compared with the saline control. In the other period of the antibody response, although DA content in these lymphoid organs had some changes, there was no marked significance. Above results show that on the one hand, DA can enhance the function of the cellular immunity and on the other hand, the humoral immunity can affect the content of DA in the lymphoid organs, which suggests that an interaction exists between the DA and immune system.
