Optimization of Extract Technology of Amygdalin in Amygdala amara from Xinjiang by Orthogonal Experiment

摘要 目的:优选新疆苦巴旦杏仁中苦杏仁苷的提取工艺。方法:以苦杏仁苷含量为定量指标,通过单因素试验优选出的微波灭酶时间、苦巴旦杏仁细粉与石油醚比例、脱脂粕与95%乙醇的比例为考察因素,用正交试验优选最佳提取工艺。结果:最佳提取工艺为微波灭酶3.5min,苦巴旦杏仁细粉与石油醚比例为1∶6进行脱脂,脱脂粕与95%乙醇为1∶30进行纯化,在此条件下苦杏仁苷的含量为3.21%。结论:该提取工艺合理、简单、可行,可为大生产提供理论依据。 OBJECTIVE: To optimize the extract technology of amygdalin in Amygdala amara from Xinjiang. METHODS: Orthogonal experiment was carried out to optimize the extraction technology with the content of amygdalin as quantitative index, and with the duration of enzyme - deactivation by microwave, the ratio of Amygdala amara fine powder to petroleum benzin, and the ratio of defatted meal to ethanol as factors. RESULTS: The optimum extraction conditions were as follows: the duration of enzyme - deactivation by microwave was 3.5 min; the ratio of Amygdala amara fine powder to petroleum benzin was 1 : 6,and the ratio of defatted meal to 95% ethanol was 1 : 30. Under the above conditions, the content of amygdaloside reached 3.21%. CONCLUSION: The extraction technology was proved to be reasonable, simple and feasible, and it provides theoretical basis for large production.
