镉对大鼠睾丸间质细胞脂质过氧化的影响 被引量:11

Effects of cadmium on lipid peroxidation of Leydig cells of rat testis in vitro

摘要 目的观察镉对离体睾丸间质细胞的毒性作用。方法测定不同CdCl2染毒浓度下(含镉0、10、20和40μmol/L)间质细胞的脂质过氧化水平。结果染毒24小时后,染毒组的细胞存活率显著低于对照组(P<0.01),且存在剂量-效应关系;随染毒浓度的增高,丙二醛(MDA)含量和谷胱甘肽过氧化物酶(GSH-Px)活力显著增高(P<0.05),而超氧化物歧化酶(SOD)活力则显著下降(P<0.01)。相关分析结果显示:SOD与MDA呈负相关、与GSH-Px呈显著负相关(r=-0.4339,P<0.05);MDA与GSH-Px呈显著正相关(r=0.4329,P<0.05),表明镉可使睾丸间质细胞的脂质过氧化作用增强、抗氧化能力减弱。结论镉对睾丸间质细胞具有直接毒性作用。 In order to study the toxic effects of cadmium on Leydig cells of rat testis,lipid peroxidation levels of Leydig cells exposed to different concentration of cadmium(i.e.0,10,20 and 40 μmol Cd/L) in vitro were determined.The results showed that twenty-four hours after exposed to cadmium,the cells' survival rate in the exposed groups was significantly lower than that in the control group(P<0.01).It was found that there were dose effect relationships between the cell survival rate and cadmium exposure.With increased cadmium exposure dose,the contents of MDA and GSHPx activity in the exposed groups were significantly higher than those in the control(P<0.05),but SOD activity was inverse(P<0.01).Correlation analysis showed that there were negative correlation between SOD and MDA or GSHPx(r=-0.433 9,P<0.05),and positive correlation between MDA and GSHPx(r=0.432 9,P<0.05).It is indicated that Cd could increase lipid peroxidation level and reduce antioxidation ability of Leydig cells from testis in rat,suggesting that cadmium may have direct toxic on them.
