摘要
目的:传染性非典型肺炎也称"严重急性呼吸综合征"(severe acute respiratory syndrome,SARS)血清快速检测试纸条的研制以及在SARS患者、发热及疑似SARS与健康人的早期鉴别诊断应用的探讨。方法:收集了13例SARS确诊患者、135例发热及疑似SARS患者、10例胸膜炎患者、9例肺癌患者、30例肺结核患者和50例健康人的血清(对照组),应用SDS-PAGE分别对上述患者、健康人血清进行电泳,应用离子交换层析法分离纯化SARS患者血清分子量为61 kDa的特种蛋白,Western blotting鉴定分离纯化的特种蛋白,氨基酸序列测定仪分析特种蛋白的氨基酸序列,酶化学法制备SARS血清快速试纸条。结果:分子量为61 kDa的特种蛋白仅存在于SARS患者血清中,研制的SARS血清快速检测试纸条的敏感性、特异性和准确度分别为92.31%、97.12%、97.00%。结论:在SARS早期鉴别诊断中,SARS血清快速试纸条的成功研制有较高的临床应用价值,有助于SARS的排除,值得临床推广应用。
Objective: To develop SARS sera rapid test strip and to explore the application of early differentiated diagnosis in patients respectively with severe pulmonary tuberculosis, suspected SARS, SARS and health. Methods:Sera were collected from 13 patients with SARS, 135 patients with suspected SARS and 50 healthy volunteers. Meanwhile all the patients with SARS, suspected SARS and health volunteers serum electrohyresized by SDS - PAGE. Moreover special protein with 61 kDa, Molecular weight was purified by ion exchange from patients with SARS in serum chromatography furthermore, the amino acid sequence of special protein was analyzed by amino omni acid sequencer . The special protein was identified by western blotting. In the end, The SARS sera rapid testing strip was developed in accordance with enzymic theory chemically. Result:Special protein with 61 kDa molecular weight is only included in SARS patients sera . The sensitivity specificity and accuracy of SARS sera rapid testing strip were respectively 92.3 %, 97.12 % , 97.00 %. Conclusion:Successful development of SARS sera rapid testing strip has marked medically certain breakthrough in the early differentiated diagnosis of SARS.
