摘要
目的建立高效液相色谱法(high performance liquid chromatography,HPLC)测定雷公藤提取物中雷公藤甲素、雷公藤精碱和wilfornine F的含量。方法采用YMC-Pack ODS-A(4.6mm×150mm,5μm)色谱柱,以乙腈-0.02mol/L磷酸二氢钠水溶液为流动相梯度洗脱,210nm波长处检测,流速1.0mL/min,温度25℃。结果雷公藤甲素、雷公藤精碱和wilfornine F在一定范围内均呈良好的线性关系,平均加样回收率在98.81%~99.84%范围内,RSD≤1.42%(n=6)。结论该方法准确、重复性好,适用于雷公藤提取物的质量评价。
Objective To develop a high performance liquid chromatography (HPLC) method for the determination of triptolide, wilforgine and wilfornine F in the extract of Tripterygium wilfordii Hook. f. Methods The HPLC separation was carried out on a YMC-Pack ODS-A column (4.6 mm ×150 mm, 5 μm) with acetonitrile - 0.02 mol/L sodium dihydrogen phosphate as the mobile phase for gradient elution. The flow rate was 1.0 mL/min, the detection wavelength was 210 nm, and the temperature of column was 25 ℃. Results Linearity was confirmed for triptolide, wilforgine and wilfornine F. The average recoveries were in the range of 98. 81%- 99. 84% with RSDs less than 1.42 % (n = 6). Conclusions The method is accurate, repeatable and applicable to the determination of triptolide, wilforgine and wilfornine F in T. wilfordii.
