地衣芽孢杆菌发酵培养基的优化 被引量:14

摘要 [目的]对地衣芽孢杆菌发酵培养基进行优化,并在此基础上确定最佳培养条件,为实现其工业化生产提供参考。[方法]借鉴谷春涛等关于地衣芽孢杆菌TS-1液体培养发酵条件的研究成果,对培养基进行澄清,并运用单因素试验和4因素3水平正交试验对其培养基进行改进,对地衣芽孢杆菌发酵条件进行优化以得到其最佳温度、pH值和接种龄。[结果]4种因素对地衣芽孢杆菌发酵影响的显著性次序为:玉米浆〉豆饼粉浸汁〉蛋白胨〉葡萄糖。培养基的最佳配比为:玉米浆0.45g,蛋白胨1.50g,豆饼粉浸汁为1:15,葡萄糖1.50g。最适宜种龄为18h,接种量8%,起始pH值7.5,最佳温度37.5℃,此条件下地衣芽孢杆菌对数生长期在14~20h,发酵周期为22~24h。活菌数每单位提高近1亿个,发酵周期比经验周期缩短10h。[结论]试验得出了地衣芽胞杆菌发酵培养基的最佳配比及最佳培养条件,在此条件下培养,收获菌体量大大提高,降低了生产成本,更有利于其大规模工业化生产。 [Objective] On the basis of the optimization of the Bacillus licheniformis fermentation medium, the best culture condition was detemtined for the reference for the implementation of its industrial production. [Method] On the basis of the results from Gu Chtmtao in the liquid culture fermentation condition of Bacillus licheniformis-TS-1, the further test of the culture medium was conducted, and the experinaent in the medium improvement for bacillus licheniformis was done with the single-factor orthogonal test with 4 factors and 3 levels for the optimum temperature, pH value and age of inoculation. [ Results ]The order of four kinds of factors significantly impacting the fermentation of Bacillus licheniformis was as follows: corn juice 〉 steep liquor from bean cake powder 〉 peptone 〉 glucose, and the best contain in the medium was 0.45 g corn steep liquor, 1.50 g peptone, the ratio of steep liquor from bean cake powder in the medium of 1 : 15 and 1.50 g glucose. The most appropriate inoculation age was 18 hours; inooalation amount, 8% ; the initial pH value, 7.5 mad the best temperature, 37.5 ~C ; under the condition, the logarithmic growth time of bacillus lichenifonnis appeared from the 14th to 20th hour, and the fermentation period was 22 - 24 hours. The number of viable cells per unit was raised to near 100 million cells and the cycle of the fermentation was shortened for more than 10 hours. [Conclusion] Tne best Bacillus licheniformis fermentation medium and culture condition were obtained, under the condition, the harvested biornass was greatly improved and the production cost was reduced, which was more conducive to its large-scale industrial production.
