大肠癌组织中HPV和C-erbB-2基因的检测及其关系研究

Detection and analysis of HPV and C-erbB-2 genes in colon cancer.

目的检测大肠癌组织中HPV和C-erbB-2基因,探讨大肠癌组织中HPV的存在与C-erbB-2基因的扩增之间的关系。方法应用PCR技术检测21例大肠癌标本中HPV及C-erbB-2基因。检测C-erbB-2基因的同时进行差异PCR,以确定C-erbB-2基因拷贝数的变化。结果 4例(19.05%)HPV 16型阳性,其中1例HPV 16、18双重阳性。6例(28.57%)C-erbB-2基因拷贝数增加。2例HPV16型阳性的同时C-erbB-2基因拷贝数增加。结论我国大肠癌组织中亦存在HPV感染现象,部分HPV感染的病例可同时存在C-erbB-2基因的扩增。

Objective To detect the human papilloma virus (HPV) and C-erbB-2 genes in colon cancer and ana-lyze the relationship between them. Methods The HPV and C-erbB-2 genes were detected in surgical specimens of 21 patients with colon cancer using PCR technique. At the time of detection of C-erbB-2 gene,the differential PCR was used to determine the changes in copies of C-erbB-2 gene. Results Of the 21 samples, 4 were HPV16 positive,including one positive both for HPV16 and HPV18, 6 showed overex-pression of C-erbB-2 gene. In the 2 cases of HPV positive, C-erbB-2 gene was overexpression at the same time. Conclusion The findings confirmed the presence of HPV infection in colon cancer. Some cases with HPV infection might simultaneously have C-erbB-2 overexpression in colon cancer.
