摘要
目的探讨益气解毒方对大鼠鼻咽癌变过程中细胞凋亡和增殖的干预作用。方法用二亚硝基哌嗪(DNP)诱导大鼠诱发鼻咽癌变模型,益气解毒方进行干预治疗。定期分批处死大鼠,取鼻咽组织做病理检测,TUNEL法检测细胞凋亡活性,免疫组织化学SABC法检测PCNA蛋白活性表达。结果益气解毒方组大鼠鼻咽上皮组织的癌变率为0%,明显低于生理盐水模型组(86.3%);维甲酸阳性对照组大鼠的癌变率为0%,但该组所有动物均于实验周期的第250~310天间逐渐死亡。在诱发性鼻咽上皮细胞癌变进程中的单纯性增生阶段,大鼠鼻咽上皮细胞凋亡活性和PCNA蛋白表达活性组间差异均无统计学意义(P>0.05);在异型增生阶段,益气解毒方组鼻咽上皮细胞凋亡活性明显高于模型组,差异具有统计学意义(P<0.05),PCNA蛋白表达活性则显著低于模型组,差异具有统计学意义(P<0.05)。结论益气解毒方能有效阻逆大鼠鼻咽上皮细胞癌变进程,其作用与其有效诱导细胞凋亡、抑制细胞增殖活性有关。
Objective To investigate the effects of Yiqi Jiedu decoction(YJD) on the apoptosis and proliferating in the developing process of carcinogenesis in rats. Mehtods The nasopharyngeal carcinoma (NPC) models were induced by N. N'-dinitrosopiperazine (DNP), treated with YJD, and killed regularly to get the nasopharyngeal tissue for pathohistological evaluation. Apoptotic activities was detected with TUNEL(TdT-mediated x-dUTP nick end labeling) and the activity expression of PCNA with SABC immunohistochemical. Results The rate of cancerously transformed nasopharyngeal epithelia was 0% in experimental group (EG) treated by YJD, which lowered significantly than that in model group (MG) (86.3%) feeding saline; the rate in control group (PCG) was also 0%, but all the animals were gradually dead from 250th to 310th day. In the simple hyperplasia stage induced cancerous transforming lesion, The differences of apoptotic index and PCNA activity between the 3 groups were not significant(P〉0.05); In the pathological stage of atypical hyperplasia, the apoptotic index in EG was significantly elevated (P〈0.05) and PCNA activity significantly declined (P〈0.05) than those in MG. Conchusions YJD is effective for anticancer, which may be concerned with that YJD can increase the activity of apoptosis and inhibit the cell proliferating.
