通塞颗粒对慢性阻塞性肺疾病急性加重期大鼠模型肺组织细胞外基质的影响 被引量:2

Effect of Tongsai Granule on the Extracellular Matrix of Lung Tissue in the Acute Exacerbation of Rats with Chronic Obstructive Pulmonary Disease

摘要 目的探讨通塞颗粒对慢性阻塞性肺疾病(COPD)急性加重期模型大鼠肺组织细胞外基质的作用。方法将Wistar大鼠随机分为7组,空白组、稳定期模型组、加重期模型组、高剂量组、低剂量组、氨茶碱组、葶贝组,每组10只,以肺炎克雷伯杆菌经鼻腔滴入法建立COPD急性加重期大鼠模型,空白组、COPD稳定期、加重期模型组以生理盐水灌胃,余组以相应剂量药物灌胃,每日2次。放免法监测血清和支气管肺泡灌洗液(BALF)中型前胶原(PC)、层粘连蛋白(LN)、透明质酸(HA)的表达变化,免疫组化法监测肺组织细胞外基质成分的变化。结果血清和BALF中PC、LN、HA表达显示加重期模型组均明显高于稳定期模型组和空白组(P<0.01),稳定期模型组均明显高于空白组(P<0.01);高剂量组均明显低于加重期模型组(P<0.05或P<0.01)。图像分析表明,各组肺组织中型、型、型胶原、LN、TGF-β1的表达均明显高于空白组(P<0.01);加重期模型组、氨茶碱组和高剂量组肺组织肺组织中型、型、型胶原、LN、TGF-β1的表达,低剂量组型、型、型胶原的表达均明显低于加重期模型组(P<0.01)。结论通塞颗粒能抑制COPD急性加重期肺组织细胞外基质成分的降解、气道结构的重塑,以通塞颗粒高剂量组作用最为明显。 Objective To study the effect of Tongsai Granule (Granule for Removing Obstruction) on the extracellular matrix (ECM) of lung tissue in the acute exacerbations of rats with Chronic Obstructive Pulmonary Disease (COPD). Methods The Wistar rats were randomized into 7 groups with 10 in each: Blank control group, stable stage model group, acute exacerbation model group, High dosage of Tongsai Granule group, Low dosage Of Tongsai Granule group, Aminofilina control group, Tingbei Capsule control group. The COPD acute exacerbation models were developed by Pneumonia Klebsiella Bacillus nasal dropping. The blank control group and model groups were given normal saline and the other groups were given corresponding medicines by gavage, twice a day. The expression of PC Ⅲ ,LN and HA in serum and bronchoalveolar lavage fluid (BALF) were monitored by radio-immunity (RI) method, and the changes in the extracellular matrix of lung tissues were detected by immunohistochemistry method. Results The expression of PC Ⅲ ,LN and HA in serum and BALF in acute exacerbation model group was significantly higher than that of stable model group (P〈0.01) and blank control group (P〈0. 01). The expression of PC Ⅲ ,LN and HA of stable model group was significantly higher than that of the blank control group (P〈0. 01). The expression of PC Ⅲ,LN and HA of high dosage of Tongsai Granule group was significantly lower than that of acute exacerbation model group (P(0.05 and P〈0.01). The image analysis showed that the expression of collagen Ⅰ , collagen Ⅲ , collagen Ⅳ , LN, TGF-β of lung tissue in all groups was significantly higher than that of the blank control group (P〈0. 01). The expression of collagen Ⅰ , collagen in , collagen Ⅳ , LN, TGF-β of lung tissue in acute exacerbation model group, aminofilina control group and high dosage of Tongsai Granule group, and the expression of collagen Ⅰ, collagen Ⅱ and collagen Ⅳ of low dosage of Tongsai Granule were both significantly lower than those of acute exacerbation model group (P〈0. 01). Conclusion Tongsai Granule has the effect to restrain the degradation of ECM in acute exacerbation of COPD and to restrain the remodeling of air passage structure, the effect of high dosage of Tongsai Granule group is the most remarkable.
