摘要
目的探讨藻黄胶囊Ⅲ号(ZHC3)治疗慢性肾功能衰竭的机理。方法采用不同浓度的ZHC3孵育体外培养的人肾间质成纤维细胞(HRIF),观察24h和48h时HRIF增殖与凋亡情况,检测纤维连接蛋白(FN)、层粘连蛋白(LN)、转化生长因子β1(TGF-β1)的分泌水平。结果各浓度的ZHC3在24h和48h时可显著抑制HRIF的增殖(P<0.01);各浓度的ZHC3在24h时减少FN的分泌(P<0.01);25μg/ml和50μg/ml浓度的ZCH3可在48h时减少TGF-β1分泌(P<0.05);对LN的分泌及HRIF的凋亡无明显影响。结论ZHC3可抑制HRIF的增殖,而且可以减少FN、TGF-β1的分泌,这可能是ZHC3治疗慢性肾功能衰竭的机理。
Objective To study the mechanism of Zaohuang Capsule No. 3 (ZHC3) on treating the chronic renal failure. Methods ZHC3 in different concentration was used to foster the cultured human renal interstitial fibroblasts (HRIF) in vitro to observe its proliferation and apoptosis and to detect the secretion of fibronectin (FN), laminin (LN), and transforming growth factor β1 (TGF-β1) at the time points of 24h and 48h. Results ZHC3 in different concentration inhibited significantly the proliferation of HRIF at 24h and 48h (P〈0. 01). ZHC3 in different concentration decreased the secretion of FN at 24h (P〈0.01). ZHC3 in 25ug/ml and 50ug/ml decreased the secretion of TGF-β1 at 48h (P〈0.05). There was no effect on the secretion of FN and apoptosis of HRIF. Conclusion ZHC3 inhibits the proliferation of HRIF and decreases the secretion of FN and TGF-β1, being possibly the mechanism for treating chronic renal failure.
