瘦素对重症急性胰腺炎大鼠肺组织NF-κ B、iNOS及NO的影响 被引量:3

Influnence of Leptin On Nuclear Factor-κ B/iNOS Activity and NO Content in the Rats Lungs with Severe Acute Pancreatitis

摘要 目的探讨外源性瘦素对重症急性胰腺炎大鼠肺组织核因子-κB、诱生型一氧化氮合酶、NO表达的影响,以及瘦素在SAP肺损伤的治疗效果及作用机制。方法36只大鼠随机分为正常对照组、SAP组和外源性瘦素治疗组。以5%牛磺胆酸钠制作SAP模型。造模后6h取材,检测肺组织NF-κB活性、NO、iNOS含量,同时观察血浆淀粉酶及其病理改变等。结果正常对照组几乎无NF-κB活化,SAP时NF-κB活化及iNOS、NO高表达。瘦素对SAP肺组织NF-κB活性有抑制作用,可使iNOS活性表达降低,并可降低血浆淀粉酶。肺病理可见:给予外源性瘦素后,SAP大鼠肺组织损伤明显减轻。肺组织NF-κB活化与iNOS表达及肺组织病理评分均呈正相关。结论肺组织NF-κB活化及iNOS过度表达可能是SAP肺损害发生的原因之一。瘦素可能通过抑制NF-κB活性途径减少iNOS的表达,从而减少过量NO的生成,减轻SAP肺组织损害。 Objective To explore the relationship between expression of iNOS,NO,NF-κB activation of lungs tissue and lungs injury in rats with SAP and the effect of leptin on SAP. Methods 36 rats were randomized into the three group:normal group,SAP group,leptin treatment group. The SAP model was induced by injection of 5% sodium taurocholate solution intobiliopancreatic duct. All the animals were sacrificed in 6h. Activation of NF-κB was determined and expression of NO was assayed. Mean-while,iNOS,plasma level of amylase,and histological grading were determined. Results In SAP group,the NF-κB activity,the NO levels and the iNOS expression increased markedly.In Leptin group, the activation of NF-κB was prevented and plasma level of amylase, and content of NO and iNOS remarkably decreased. Conclusion Early blockage of NF-κB activation with leptin is effective for reducing severity of the lungs injury.Leptin could reduce iNOS expression and NO level by inhibiting the NF-κB activation. Leptin may be beneficial marker for SAP.
