两个蓝莓品种离体叶片不定芽再生体系的建立 被引量:20

摘要 以Sunrise和Geo两个蓝莓品种试管苗叶片为外植体材料,以改良的WPM为基本培养基,研究了暗培养时间、叶片放置方式和不同浓度激素组合(ZT、IBA、TDZ)对2个蓝莓品种叶片不定芽再生的影响。结果表明:最适的暗培养时间为14 d,近轴面放置效果最好,不同基因型不定芽再生能力不同,Sunrise品种在WPM+4.0 mg/L ZT+0.3 mg/L IBA+1.0 mg/L TDZ+20 g/L蔗糖+6 g/L琼脂培养基中生长最好,离体叶片不定芽再生频率达100%,再生不定芽数可达4.5个以上;Geo品种最适宜培养基为WPM+4.0 mg/L ZT+0.3 mg/L IBA+20 g/L蔗糖+6 g/L琼脂,叶片不定芽再生频率达到100%。 With the two blueberry varieties "Sunrise" and "Geo" as experimental materials and the improved WPM as basic medium, the effects of dark incubation time, way of leaf touching medium and different hormone combinations on adventitious shoot regeneration from detached blueberry leaves were studied. The results were as follows: the optimum dark incubation time was 14 days, the adaxial leaf touching medium was better. The regeneration abilities of different blueberry varieties were different. The optimum medium for "Sunrise" was WPM + 4.0 mg/L ZT + 0.3 mg/L IBA + 1.0 mg/L TDZ + 20 g/L sucrose + 6 g/L agar with the regeneration frequency of 100% and more than 4.5 adventitious shoots; the optimum medium for "Geo" was WPM + 4.0 mg/L ZT + 0.3 mg/L IBA + 20 g/L Sucrose + 6 g/L agar with the regeneration frequency of 100%.
