摘要
目的:探讨IL-1β对神经元样PC12细胞IL-1RⅠ蛋白表达的影响。方法:将PC12细胞传代后以一定的密度种植于培养皿后,NGF诱导其分化成神经元样细胞后,随机分为空白对照组(A组)I、L-1β1ng/mL刺激组(B组)I、L-1β5ng/mL刺激组(C组)、IL-1β10ng/mL刺激组(D组)。24h后分别以MTT法测定各组细胞存活率,Western Blotting方法检测神经元样PC12细胞内IL-1RⅠ蛋白表达。结果:MTT法显示,与A组相比,不同剂量组的IL-1β均可导致神经元样PC12细胞存活率的下降(P<0.01);WesternBlotting方法显示,与A组相比,不同剂量组的IL-1β刺激均可引起神经元样PC12的IL-1RⅠ蛋白表达上调(P<0.05)。结论:IL-1β可引起神经元样PC12细胞的IL-1RⅠ蛋白高表达,这可能与其所致神经元样PC12细胞细胞存活率的下降相关。
Objective: To investigate the interfering effect of IL-1β on the expression of IL-1R Ⅰ protein in neuronal PC12 cells. Methods: PC12 cells were seeded and exposed to 7S-NGF after 24h which could induce PC12 cells to differentitate into neuronal cells, then were randomly allocated to group A(control group), group B (1ng/ml IL-1β-induced group), group C (5ng/ml IL-1β-induced group), group D (10ng/ml IL-1β- induced group). Cell viability was monitored by the colorimetric MTT assay, the expression of IL-1R Ⅰ protein was tested by the Western blot analysis after IL-1β induced 24h. Results: Cell viability of group D, was significantly lower than that of group A, group B, group C (P〈0.01). The expression of IL-1R Ⅰ protein of group Dwas significantly higher than that of group A, group B, group C(P〈0. 01). Conclusion: IL-1β may cause the higher expression of IL-1R Ⅰ protein on neuronal PC12 cells,which is related to the neuronal PC12 cell damage induced by IL-1β.
