瑞芬太尼对大鼠肠缺血再灌注损伤时肝细胞凋亡及caspase-3的影响 被引量:1

摘要 目的探讨瑞芬太尼对大鼠肠缺血再灌注损伤时肝细胞凋亡及caspase-3的影响。方法成年健康Wistar大鼠42只,随机分为3组:对照组(C组,n=6)、肠缺血再灌注组(IR组,n=18)、瑞芬太尼干预组(R组,n=18)。各组根据再灌注时间分为1,3,6h三个时相。通过夹闭肠系膜上动脉制作肠缺血再灌注损伤模型,实验结束后即刻取肝左叶做标本。用免疫组化法检测caspase-3蛋白表达量,TUNEL法检测肝细胞凋亡指数(AI)。结果与C组比较,IR组和R组肝组织caspase-3蛋白含量、AI均增加(P<0.05);与IR组比较,R组caspase-3蛋白表达和AI减少(P<0.05)。结论瑞芬太尼可以下调大鼠肠缺血再灌注时caspase-3蛋白表达,使肝细胞凋亡减轻,从而对肝损伤有一定的保护作用。 Objective To investigate the effects of remifentanil on hepatic cell apoptosis and expression of caspase-3 in rats with intestinal ischemia-reperfusion injury. Methods Forty-two Wistar rats were randomly divided into 3 groups : control group ( C group, n = 6) ,ischemia-reperfusion group( IR group, n = 18 ), remifentanil group (R group, n = 18 ). The rats were executed in each group after reperfusion for 1 h,3 h and 6 h, respectively. A rat model of intestinal ischemia-reperfusion was established by blocking the superior mesenteric artery with clap. The left hepatic lobes were sampled at the end of reperfusion. Expression of caspase-3 protein in liver tissues was assessed by immunohistochemical method. The apoptosis index (AI)of hepatic cells was measured by TUNEL method. Results Caspase-3 protein expression and AI of hepatic cells were significantly higher in IR group and R group than in C group( P 〈 0.05 ) , but lower in R group than in IR group ( P 〈 0.05 ). Conclusion Remifentanil can relieve the hepatic cell apoptosis by decreasing caspase-3 protein expression,which may protect liver from intestinal isehemia-reperfusion injury.
