摘要
目的观察蛋白激酶C-α(PKC-α)mRNA在兔自体静脉移植中的表达的动态变化,探讨其与移植血管内膜增生的关系。方法采用兔自体颈静脉移植模型(右颈静脉移植至对侧颈动脉),25只新西兰大白兔随机分为5组(n=5),为Ⅰ组、Ⅱ组、Ⅲ组、Ⅳ组、SO组。Ⅰ-Ⅳ组的取材时间分别为移植术后的第3,7,14,28天。SO组只行颈部切开与再缝合,未行血管移植,术后直接取材。应用real-time PCR法检测各组中PKC-α mRNA的表达变化。结果与SO组相比,PKC-α mRNA在静脉移植术后第3天时,表达升高至2.335±0.059倍(P<0.01),而后在术后7,14,28d表达渐降至正常。结论PKC-α可能与自体静脉移植后的血管内膜增生过程中的信号传导有关。
Objective To explore the expression of PKC-α mRNA in autogenous vein graft and its relationship with intimal hyperplasia. Methods Twenty-five New Zealand rabbits were randomized into five groups:group Ⅰ , group Ⅱ , group m, group Ⅳ and SO group. The right common jugular vein was transplanted to the contralateral common artery in group Ⅰ - Ⅳ. Then the specimen was collected after transplantation for 3,7, 14,28 d in group Ⅰ, group Ⅱ, group Ⅲ group Ⅳ, respectively. The rabbits were sham-operated in SO group, and then the specimen was immediately collected after operation. The PKC-α mRNA was detected by real-time PCR. Results Compared with SO group, the PKC-α mRNA expression increased 2. 335 ± 0. 059 fold after transplantation for 3 d( P 〈 0.01 )and then declined gradually. Conclusion PKC-α might be involved in the signal transduction of the smooth muscle cell proliferation in autogenous vein graft.
