摘要 目的:探讨增殖细胞核抗原(Ki67)在绒毛膜癌5-氟尿嘧啶(5-FU)治疗中的作用.方法:用绒毛膜癌JAR细胞株进行培养传代,以不同浓度5-FU作用于JAR细胞,MTT方法观察5-FU对JAR细胞生长的抑制情况,用逆转录聚合酶链(RT-PCR)和免疫印迹技术(Western-Blot)方法检测Ki67基因和蛋白的表达.结果:①5-FU对JAR细胞的生长具有抑制作用,并有浓度和时间的依赖性;②5-FU作用于JAR细胞后Ki67基因和蛋白表达减少,随浓度增加而减少.结论:Ki67可以指导绒毛膜癌5-FU的化疗;也可以反映绒毛膜癌5-FU化疗的效果. AIM: To investigate the role of proliferating cell nuclear antigen(Ki67) in chemotheraphy by 5-FU in choriocarcinoma. METHODS:Choriocarcinoma cell JAR strain was used in this study and the inhibitory effects of chemotherapeutical drug 5-Fu (5-Fluorouracil) on JAR strain were assayed with MTF method. Expression of Ki67 was detected by reverse transcriptase PCR and Western blot. RESUTLS:5-FU inhibited the JAR cell growth in a close and time dependent manner. Ki67 gene and protein decreased with the increase of 5-Fu. CONCLUSION: Ki67 is an important index in chemotheraphy by 5-FU and reflects the effect of chemotheraphy by 5-FU on choriocarcinoma.
