Expression and distribution of semphorin 3f(SEMA3F) and its receptor neuropilin-1 and neuropilin-2 in oral carcinoma and their potential function

摘要 目的研究SEMA3F及其受体Nrp1和Nrp2在口腔癌组织和细胞(ACC2,ACCM和TCa)中的表达情况,并推测其在口腔癌生成中的可能意义。方法应用RT-PCR和Western bloting以及免疫组化法检测SEMA3F、Nrp1及Nrp2在口腔癌(ACC2,ACCMand TCa)组织和细胞中的表达情况。结果在所有口腔癌组织和癌细胞中均可检测到Nrp1和Nrp2的表达,但Nrp2相对表达较弱。Western bolting、RT-PCR提示Nrp1呈高表达,Nrp2及SEMA3F均表达较低。在免疫组织化学中显示癌及癌周组织中Nrp1均呈高表达,且癌巢中的表达明显高于癌间隙组织,Nrp2及SEMA3F呈较弱表达。结论提示在肿瘤的发展进程中,Nrp1可能对肿瘤本身的发生,增殖及肿瘤的血管生成均可能起到了重要的作用。 Objective To investigate the expression and distribution of the semphorin 3f(SEMA3F)and its receptor neuropilin-1 and neumpilin-2 in the tissue and the cells of the head and neck carcinoma(ACC2, ACCM and TCa) and explore their potential function in the tumor. Methods Reverse transcription polymerase chain reaction(RT-PCR) and the western blotting were used to survey the expression of SEMA 3F, neuropilin-1 and nearopilin-2 with specific primers and antibody. Immunohistochemistry staining for sema3F, neuropilin-! and neuropilin-2 were also performed in the tissues. Results Neuropilin-l,2 mRNA were detected in all the oral carcinoma tissues and the cells, but very weak signals for neuropilin-2, Sema3F were detected. Western blotting indicated that the high expression of the NRP-1, the low expression of the neurepilin-2, SEMA 3F. Immunohistochemistry indicated that NRP1 was highly expressed in the tissue of the carcinoma, especially in the cancer nest. Conclusion The high expression of NRP1 indicated that NRP1 may play an importmant part in the process of the tumour gener- ation, both in the tumor proliferation and in the angiogenesis.
