4,5,7-三羟基异黄酮和阿霉素诱导人舌癌细胞株Tca8113凋亡的研究 被引量:1

摘要 目的观察4,5,7-三羟基异黄酮(Genistein)和阿霉素(ADM)单用和联用对诱导体外培养的人舌癌细胞株Tca8113凋亡的影响。方法以体外培养的人舌癌细胞株Tca8113为实验模型,Genistein与ADM联合作用于癌细胞,倒置相差显微镜下观察药物处理后癌细胞的细胞形态学变化,四甲基偶氮唑蓝比色法分析药物对癌细胞的增殖抑制作用,流式细胞仪测定细胞凋亡率和细胞周期分布。结果Genistein和ADM两者单用对癌细胞均有生长抑制作用,且随剂量升高抑制趋势增强(P<0.05)。两药联用对癌细胞的抑制作用优于两者的单纯相加(Q>1)。联合用药使肿瘤细胞S期延长,G2/G1值缩小,凋亡率明显升高(P<0.05)。结论Genistein与ADM联用对Tca8113细胞具有协同抑制作用,其作用机理可能是联合用药促使肿瘤细胞S期延长,G2/G1值缩小,提高了ADM的诱导凋亡效应。 Objective To observe the effects of Adramycin on the apoptosis of Tca8113 cells combined with or without Genistein. Methods TcaS113 cells were cultured in the medium containing Adramycin of different concentrations with or without Genistein. Apoptotic morphology of Tca8113 cells was observed under inverted phase contrast microseope; the effect was determined by methylthiazolyl tetrazolium(MTT)assay of Tca8113 cells;and the apoptosis of TcaS113 cells was examined by flow cytometer. Results Genistein and Adriamycin inhibited the growth of TcaS113 cells in a dose-dependent pattern( P 〈 0.05), and the combination of them showed better inhibitive effects on TcaS113 cells, the cell cycle was arrested in S phase, G2/G1 value decreased( P 〈 0.05), and the apoptosis of Tca8113 cells increased significantly( P 〈 0.05). Con- clusions Adriamycin with Genistein had synergistic effects on cytotoxicity, effectively inhibited the growth of TcaS113 cells, changed the distribution of cell cycle and induced apoptosis of Tca8113 cells. It might result from arresting in S phase.
