摘要 目的:探寻正常皮肤与增生性瘢痕中表皮干细胞标记物的表达,从组织学角度探讨表皮干细胞参与瘢痕增生的证据。方法:术中切取8例伤后6月增生性瘢痕患者的瘢痕组织及同例患者的正常皮肤,将其制备切片并分组,用Elivision二步法免疫组织化学检测表皮干细胞表面标志物β1整合素和CK19的表达,计数阳性细胞并进行统计学处理。结果:瘢痕组织真皮层中,表达阳性细胞数β1整合素和CK19较正常皮肤β1整合素和CK19明显增多(P<0.05),成纤维细胞数显著增多。结论:在病理性瘢痕的发生中,表皮干细胞可能在细胞因子作用下分化为成纤维细胞,从而参与瘢痕形成。 Objective To investigate the expression of fibroblast-like epidermal stem cells by the different location and the expression characteristics of epidermal stem cells in normal adult skin and scar tissue. Methods 8 skin tissue specimens were taken from the operated patients. As the biochemical markers for epidermal stem cells,J51 integrin and keratin 19 (K19) were employed as the first antibody and determined by Elivision two step immunohistochemistry. Results The expression of β1 integrin and the CK19 positive cell number in the epithelial basal layers of scar tissue were obviously decreased and weakened than those in normal adult skin, but were increased in dermal layers. The differentiation process of scar epidermal stem cells was different from that of normal skin (P〈0.05). Conclusion The results indicated that some epidermal stem cells may transdifferentiate into fibroblasts and play an important role during the formation of scar.
